Project title: The effects of chronic pain on the glial response in the brain
This project will be investigating the glial response in the rat brain 5 months after nerve injury. Chronic pain causes long-lasting changes in brain structure and function that is not well characterized at this stage. Theories include that chronic nerve damage causes a neuroinflammatory response in the brain, which may be mediated by glial cells that are proliferating and impacting upon structure and function.
During the course of this project, brain tissue that has been paraffin-embedded and sectioned will be immunohistochemically stained for GFAP (Glial fibrilliary acidic protein), a marker for astrocytes (a type of glia). Fluorescent and DAB (diaminobenzidine) visualization will be performed, and glial staining localized and quantified in regions of interest in the brain, including prefrontal, motor, sensory and limbic cortices. Image processing will be performed using ImagePro software, and cell counts and intensity levels quantified. Statistical analysis will be performed using GraphPad Prism software, and results incorporated into a manuscript for publishing.
Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.
Grading scheme (The final report must be worth at least 50% of final grade): 40% Lab component, 60% report.
