Project title: Interactions between the TBC-2 RabGAP and Rac GTPases
Project description: TBC-2 is a Rab5 GAP that localizes to late endosomes to regulate endosome maturation. The mechanism regulating TBC-2 membrane localization is not known. TBC-2 has been found to interact with the CED-10 Rac1 GTPase. We hypothesize that CED-10 might recruit TBC-2 to late endosomes in a nucleotide dependent manner. The student will determine if the physical interaction between CED-10 and TBC-2 is nucleotide dependent. The student will express and purify GST tagged CED-10 proteins (wild-type and mutant) to perform pull-down assays, in the presence and absence of nucleotide analogs, using C. elegans lysates containing GFP::TBC-2. The student will use western blot analysis with anti-GFP antibodies to test for interactions between GST-CED-10 and GFP::TBC-2 under different conditions. In addition the student will use ced-10 RNAi and ced-10 mutants to test if ced-10 is required for membrane localization of GFP::TBC-2. To do this the student will construct a ced-10(-); GFP::TBC-2 strain and analyze it and control strains by confocal microscopy. These experiments will determine the in vivo importance of the CED-10-TBC-2 interactions and identify potential novel mechanisms by which Rac GTPases can regulate vesicular trafficking.
Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.
Grading scheme (The final report must be worth at least 50% of final grade): 50% on laboratory performance, 50% on a final written research report of at least 6 pages and a 30 minute final oral presentation.
