Project title: Bacterial strains with defective chromosome replication protein binding sites
Project description: The Caulobacter crescentus chromosome replication origin has one critical binding site for the DnaA replication protein. Conspicuous DNA mutations at this 'DnaA box' abolish DnaA protein binding, these mutations prevent replication and they can not be placed by homologous recombination at their natural place on the chrosmome. This project has four phases: First, less server DnaA box changes will be made by a standard site-directed mutagenesis protocol. Second, these mutations will be tested for replication and recombination with the chromosome. The third phase will test if these mutations can be exchanged with the wild type binding sites. This standard procedure will therefore test if it is possible to create (as in the title) Caulobacter 'strains with defective chromosome replication protein binding sites.' The fourth phase will test the cell cycle consequences and apply these strains to find genetic suppressors and presumably novel replication control proteins. This project will be considered satisfactory if phases one and two are completed and successful if phase three is also started and shows preliminary results.
Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.
Grading scheme (The final report must be worth at least 50% of final grade): Final grade shall be based on laboratory performance as evaluated by the research supervisor (50%) and the final written research report (minimum 10 pages) graded by the supervisor and the course coordinator or the coordinator's delegate (50%).
Project status - This project is: Taken; however students may contact the professor to discuss other possible '396' projects this term.
