Project title: Recombinant expression of GFP-tagged fibrillin-1

Project description: Fibrillins are the major ~350 kDa components of extracellular microfibrils found in many tissues including blood vessels, bone and the eye. Mutations in fibrillin-1 lead to a number of heritable connective tissue disorders including Marfan syndrome, stiff skin syndrome and others. Assembly of fibrillin-1 into microfibrils is frequently compromised in these disorders.

The goal of this project is to generate a recombinant fibrillin-1 with a green fluorescent protein label that can be used to assess fibrillin-1 assembly in cell culture with live imaging approaches. The student will design cloning strategies and perform the actual cloning of expression plasmids. The generated plasmids will be transfected in various cells and recombinant expression of the GFP-tagged fibrillin-1 will be validated with antibodies and fluorescence microscopy. Live cell imaging will follow.

Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.

Grading scheme (The final report must be worth at least 50% of final grade): Final grade shall be based on laboratory performance as evaluated by the research supervisor (50%) and the final written research report (minimum 10 pages) graded by the supervisor and the course coordinator or the coordinator's delegate (50%)
