Project title: Overexpression of Claudin-1 during Gastrulation in Chick Embryos
Project description: During gastrulation, the tight junction protein Claudin-1 is highly expressed in the epiblast but its expression is turned off in those cells that ingress through the primitive streak during gastrulation to give rise to the mesoderm. We hypothesize that the downregulation of Claudin-1 expression in the ingressing cells is critical for gastrulation to proceed. The student will test this hypothesis by overexpressing Claudin-1 in chick in the epiblast so that its expression is maintained in the cells that ingress. Expression of Claudin-1 in the mesoderm will be analyzed by in situ hybridization and immunohistochemistry. Embryos will be assessed 6-24 hours later for changes in their gross morphology. Their developmental stage, length, and number of somites will be determined. Finally, embryos will be sectioned and analyzed histologically.
Prerequisite: 1 term completed at McGill + CGPA ≥ 3.0; or permission of instructor.
