Project title: Secretion of fibrillin-1 mutants
Project description: Mutations in fibrillin-1 cause the connective tissue disorder Marfan syndrome. The student will analyze a range of existing fibrillin-1 mutation constructs in cell culture assays. The goal is to determine whether the mutations cause a secretion block of the proteins. First, the existing mutation cell clones prepared with embryonic kidney cells will be analyzed. In addition, the plasmid DNA will be transfected into another cell line, MSU1.1, a fibroblast-derived cell line which is more relevant for the intended secretion studies, since fibrillin-1 is secreted from fibroblasts in vivo.
Prerequisite: 1 term completed at McGill + CGPA 3.0 or higher; or permission of instructor.
