Project title: Functional analysis of T cell responses in patients of type 1 diabetes with genetic variations in the cd25 gene
Project description: Genetic variations in the cd25 gene have been proposed as an important component in the genetic susceptibility to type 1 diabetes (T1D). It is currently unknown how these genetic determinants impact the generation of protective FOXP3+ regulatory (Treg) or pathogenic, diabetogenic T cell responses. Overall, the project will consist of studying the effects of known cd25 SNPs on the development and function of regulatory and effector T cells from healthy control and patients. One aspect of the project entails characterization of CD25 up-regulation/signalling induced by IL-2 and subsequent T cell activation in human peripheral blood mononuclear cells by means of multi- parametric, flow cytometry. Relevant signalling molecule expression, such as phosphoSTAT5, will also be assessed in these assays, providing insights into the mechanics of CD25 regulation and activity in responding T cell subsets. Information obtained from these experiments will be used to study the phenotypic and functional consequences of a SNP in the regulatory portion of the cd25 gene of patients suffering from T1D.

Grading scheme (The final report must be worth at least 50% of final grade): Final grade shall be based on laboratory performance as evaluated by the research supervisor (50%) and the final written research report (minimum 10 pages) graded by the supervisor and the course coordinator or the coordinator's delegate (50%).
