 : Benjamin Rich is a pharmacometrics consultant at Certara. He uses modeli
 ng and simulation to help clients\, from small biotech startups to large p
 harmaceutical companies\, with strategic decision making and model-informe
 d drug development\, most notably in the areas of oncology and rare diseas
 es. He obtained his PhD in Biostatistics from McGill University in 2012.Ph
 armacometrics is an interdisciplinary field at the crossroads of quantitat
 ive clinical pharmacology\, statistics and mathematical modeling. It is in
 creasingly recognized as an important component of the drug development an
 d regulatory approval process. In contrast to the more empirical models ty
 pically used in epidemiology\, for instance\, pharmacometric models tend t
 o be based on pharmacological or physiological principals and are often de
 scribed as “semimechanistic”. At the core of phamacometrics is the charact
 erization of drug concentration time-course by a type of nonlinear mixed-e
 ffects model referred to as a “population pharmacokinetic” model\, from wh
 ich various continuous exposure metrics can be derived. The model can help
  to identify sources of variability that could have an impact of dosing re
 commendations (e.g. renal impairment)\, and can be leveraged in simulation
 s to inform dose selection in special populations\, e.g. pediatrics. Ultim
 ately\, the goal is to link drug exposure to biochemical and clinical resp
 onses\, both desired and undesired\, to elucidate the drug’s risk-benefit 
 profile\, make go/no go decisions\, design future trials and select an opt
 imal dosing regimen. In this talk I will give an overview of pharmacometri
 c concepts and present several examples.\n
