Project title: Real time measurement of airway surface liquid composition of respiratory epithelial cells

Project description (50-100 words suggested): Impaired functional expression of CFTR alters the composition of the airway surface liquid, a phenomenon implicated in the pathogenesis of cystic fibrosis. We plan to set up model to compare the pH, chloride and glutathione concentrations in real-time, using wild type and delta508 CFTR expressing polarized human respiratory epithelial cell on filter support under physiological conditions.  This will be accomplished by engineering various fluorescence protein sensors (ClopHensor and Grx1-roGFP2) that are specifically targeted to the extracellular surface of the apical plasma membrane. Epithelial cells will be transduced by lentivirus and the fluorescence will be monitored by laser confocal fluorescence microscopy.  The effect of various metabolic conditions and CFTR channel activation will also be examined.

Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.

Grading scheme (The final report must be worth at least 50% of final grade): Final grade shall be based on an evaluation of laboratory (or equivalent) performance (50%) and a final written report (50%)  by the supervisor.
