Project title: Effect of low dose methotrexate on human hematopoietic stem and progenitor cells

Project description (50-100 words suggested): This project will evaluate the effect of methotrexate (MTX), the most commonly used drug modifier of the activity of rheumatic diseases (DMARD) in the treatment of rheumatoid arthritis, on a very specific group of cells: hematopoietic progenitors and stem cells (HSPC) that are responsible for the generation of all the mature cells of the immune system. HSPC that normally circulate in the blood reflect the function of those in the bone marrow. We have shown that HSPC from RA patients have a defect in their function. Importantly we also observed that in those patients treated with methotrexate, the function of HSPC is partially restored.

This project will define: 1- What is the effect of MTX on the function of HSPC; 2- What are potential mechanisms involved in the beneficial effect of MTX on RA-HSPC.

Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.

Grading scheme (The final report must be worth at least 50% of final grade): Final grade shall be based on laboratory performance as evaluated by the research supervisor (50%) and the final written research report (minimum 10 pages) graded by the supervisor and the course coordinator or the coordinator's delegate (50%).

Other project information: Specific objectives have been discussed with the student (literature review, experimental design-performance and interpretation, integration of new data to existing information). The evaluation will be based on the accomplishment of these points.
