Project title: Viability Studies on HeLa and HeLa-R in the Presence of Green Tea Catechins

Project description:

The principle objective of this summer project is the investigation of the influence of green tea catechins on the viability of adenocarcinoma cervical cancer cells (HeLa) and a multidrug resistant variant (HeLa-R). Several catechins are known for their chemopreventive and therapeutic effects on cancer by inducing apoptosis, inhibiting proliferation and functioning as an antioxidant in a wide range of tumor cell-lines. These proposed preliminary studies will allow further detailed metabolic studies of cancer cells in the presence of green tea catechins by electrochemical methods, such as Scanning Electrochemical Microscopy.

HeLa and HeLa-R cells will be incubated in culture medium containing different concentrations of 4 catechins, namely Epicatechin (EC), Epigallocatechin (EGC), Epicatechin-3-gallate (ECG) and Epigallocatechin-3-gallate (EGCG). Concentrations ranging from 5 uM to 100 uM will be tested over incubation periods of 2 h, 4 h, 6 h and 24 h. Cell viability will be evaluated using a cell counting method and Trypan Blue staining. Thorough statistical analysis will be performed in order to find a suitable catechin concentration that allows observation of metabolic changes in cancer cells without causing cell death.

Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.

Grading scheme (The final report must be worth at least 50% of final grade): Laboratory Performance 40%; 2 Oral Presentations during Research Group Meetings 10%; Final Report 50%.
