Project title: Photobleaching rates and pulsed light
Project description (50-100 words suggested): It has recently been shown that using pulsed laser light reduced photobleaching and phototoxicity in fluorescence microscopy and live cell imaging. This has the benefit of preserving fluorescence during light intensive confocal laser scanning microscope (CLSM) imaging. It also has the benefit of preserving living samples during live cell timelapse imaging. This project will test the dependence of photobleaching of fixed enhanced green fluorescent protein (EGFP) labelled cells with different scanning configurations on the CLSM. The student will learn how to use the confocal microscope, collect images and perform image processing and analysis to extract bleach rates.
Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.
Grading scheme (The final report must be worth at least 50% of final grade): 50% for the laboratory work and 50% for the written report.
