Project title: Initation of polyomavirus DNA replication
Project description: In this project the student will investigate the mechanims by which the large T antigen (LTA) helicase of simian virus 40 (SV40) assembles at the viral origin of DNA replication in vivo. This project will make use of cellular, luciferase-based, SV40 DNA replication assay (Fradet-Turcotte et al., 2010, Virology, 399:65-76). Mutations will be introduced in the origin of repication to test the requirment of specfic DNA elements. Similalry, LTA mutant proteins will be created. The ability of these mutant origins and LTA proteins to support viral DNA replication will then be tested in transfected cells.
Prerequisite: 1 term completed at McGill + CGPA of 3.0 or higher; or permission of instructor.
Grading scheme (The final report must be worth at least 50% of final grade): 50% research report paper; 50% laboratory research performance.
