Project title:

Generation of Dominant-Negative Mutants in p66ShcA

Project description:

The ShcA adaptor protein signals downstream of receptor tyrosine kinases to induce mitogenic and tumorigenic signals in breast cancer cells. Interestingly, the ShcA gene encodes three isoforms (p46,p52,p62)arising from alternative splicing or translational initation of a downstream methionine residue. While some studies suggest that the p66 isoform can confer pro-tumorigenic properties, others suggest that it actually induces pro-apoptotic signals through the production of reactive oxygen species. We have generated breast cancer cell lines that either express p46/p52 or the p46/p52/p66 ShcA isoforms, which will be used to interrogate the importance of the p66 isoform during mammary tumorigenesis in vivo. Preliminary data suggests that p66 confers anti-tumorigenic properties in breast cancer cells. In this BIOC396 project, the student will generate a series of dominant negative mutants within p66ShcA, so that we may identify the regions within this molecule that contribute to its ability to regulate mammary tumorigenesis.
