Project title: Prostaglandin E2-mediated positive feedback regulation of cyclooxygenase-2 gene expression
Project description: The focus of the research is on elucidating the molecular factors and signaling mechanisms implicated in prostaglandin E2 (PGE2)-mediated gene regulation in inflammatory contexts. We have established that PGE2 regulates the expression of many genes, including that of the rate-limiting enzyme implicated in its biosynthesis (i.e. cyclooxygenase-2; COX-2), at the post-transcriptional/translational levels. To date, we have established that overexpression of the RNA binding protein tristetraprolin (TTP) potently reduces COX-2 mRNA stability/translation and that TTP's subcellular localization and activity are modulated by PGE2.
The assigned research project will consist of using basic molecular and cellular biology techniques (i.e. cell culture, gene silencing by shRNA, western blotting) to silence endogenous TTP expression and analyze the effects on exogenous (i.e. transfected) and endogenous COX-2 mRNA stability/translation.
Prerequisite: 1 term completed at McGill + CGPA ≥ 3.0; or permission of instructor.
