Research:

1. Studies of post absorptive gluconeogenesis using deuterated water and the kinetics of protein metabolism using ¹³C-leucine during hyperinsulinemic clamps, are done in obese subjects with type 2 diabetes in settings of uncontrolled diabetes (hyperglycaemic) or normoglycemic with amino acids at postabsorptive or postprandial levels. These studies aim at assessing if relative insulin insufficiency affects whole body protein metabolism and the utilization of endogenous and/or dietary protein. The studies include admission to hospital for 1 week with protein kinetic evaluation done on a weight maintenance diet with or without therapy and at low or sufficient dietary protein intake.
2. Studies of the cellular mechanisms of protein synthesis and breakdown during fasting and in a postprandial state, at the skeletal muscle level using hyperinsulinemic clamps and muscle biopsies. The effects of adiposity, hyperglycemia and hyperaminoacidemia on cellular mechanisms are assessed concurrently with those on whole-body glucose and leucine kinetics, in type 2 diabetes.
